Fluorescence in situ hybridization Method of operation

1. Pre-experimental cell processing

This includes collection of cells, fixation, preparation and preservation of cell suspensions.


2. Slide preparation

The prepared cell sample is dropped onto a glass slide, and the slide is treated with a solvent such as ethanol.


3. Pre-denaturation and denaturation

The probe was dropped onto the specimen slide, carefully covered with a cover slip and sealed, and the sealed slide was denatured on a hot plate at about 75 ° C for 5 minutes.


4.Hybrid

The slides were placed in a dark, humid box and placed in a constant temperature environment of about 37 ° C for about 12 hours.


5. Washing tablets

Carefully remove the coverslip and sealant, soak and dry in a water bath at about 75 ° C, add 4,6-mercapto-2-phenylindole (DAPI), cover the coverslip, and protect from light. It was observed with a fluorescence microscope for about 10 minutes.
